HPLC and mass spectrometry: what each technique actually measures

Purity and identity are separate questions answered by separate instruments. Understanding the difference makes supplier claims much easier to audit.

HPLC answers 'how much of this is one thing?'

Reverse-phase high-performance liquid chromatography separates a sample by hydrophobicity. The detector records everything that elutes, and purity is expressed as the area of the main peak relative to total peak area.

It is an excellent purity measure and a poor identity measure — a structurally similar impurity can co-elute and inflate the number.

Mass spectrometry answers 'what is this?'

Mass spectrometry ionises the sample and measures mass-to-charge ratio, giving a molecular mass that can be compared against the theoretical mass calculated from the sequence.

Agreement between observed and theoretical mass is the strongest routine evidence that the vial contains the sequence claimed on the label.

Why both are mandatory
