Serial versus individual preparation
Preparing each concentration independently from the stock vial requires more material and more weighing. Serial dilution from a single accurately prepared stock is far more economical.
Errors compound
The trade-off is that every volumetric error propagates down the series. A 3% error at the first step is still present at the last. Verify your stock concentration carefully and use accurate delivery at each transfer.
Adsorption at the low end
At the dilute end of a series, surface adsorption becomes a meaningful loss. Low-bind consumables and a carrier protein where the assay permits both help keep the bottom of your curve honest.
